The concentration of free testosterone is derived from a mathematical equation using total testosterone, sex hormone binding globulin and albumin.

Calculated free testosterone values are reliable in most clinical situations, but should not be relied upon in situations with potential massive interference by steroids binding to sex hormone binding globulin e.g. in women during pregnancy, in men during treatment inducing high levels of DHT (dihydrotestosterone)(e.g. transdermal DHT, oral testosterone) or Mesterolone.
